Aura Biosciences Refocuses on Ocular Oncology Programs and Cuts Workforce by 20%

Aura Biosciences is restructuring its operations under recently appointed Chief Executive Officer Natalie Holles, shifting its focus toward ocular oncology while reducing its workforce by approximately 20% and making multiple changes to its executive leadership team.

Holles, who assumed the CEO role in April, inherited a company developing precision therapies for solid tumors. At the time, Aura was advancing its lead candidate, belzupacap sarotalocan (bel-sar), through a Phase 3 study in adults with early choroidal melanoma while also conducting earlier-stage clinical studies in other disease areas, including non-muscle invasive bladder cancer (NMIBC).

Company Shifts Resources Toward Eye Cancer Programs

After taking over as CEO, Holles repositioned Aura as an ocular oncology company. While the company reported what it described as an encouraging early clinical profile for bel-sar in an NMIBC study, it has decided to reduce investment in that program and concentrate resources on eye cancer indications.

Aura plans to continue collecting data in its Phase 1b/2 NMIBC trial through the protocol-defined 12-month follow-up period. According to the company, this approach will preserve future strategic options while allowing it to direct resources toward ocular oncology development programs.

The company is now prioritizing bel-sar in eye cancer indications, including metastases to the choroid and cancers affecting the ocular surface. Aura said that funds made available through the reduced focus on NMIBC will be used to support “more robust clinical data generation” for ocular oncology indications.

Enrollment Completed in Phase 3 Choroidal Melanoma Study

A key development for Aura’s lead program is the completion of patient enrollment in the Phase 3 CoMpass trial evaluating bel-sar in early choroidal melanoma. Aura had previously described the study as evaluating bel-sar as a vision-sparing therapy in adults with the disease.

With enrollment complete, the company remains on track to report topline results from the trial’s 15-month primary endpoint in the second half of 2027.

Workforce Reduction and Organizational Realignment

As part of the restructuring, Aura is reducing its workforce by about 20%. The company had 113 full-time employees as of Feb. 28, meaning the reduction could affect approximately 23 employees.

Aura disclosed that costs and cash payments associated with the workforce reduction and organizational realignment are expected to range between $2.9 million and $3.2 million, with most of those expenses anticipated during the third quarter.

The company stated that the workforce reduction “resulted in the loss of longer-term employees, the loss of institutional knowledge and expertise, and the reallocation and combination of certain roles and responsibilities across the organization, all of which could adversely affect our operations.”

Executive Team Changes Accompany Strategic Shift

The restructuring also includes several changes within Aura’s executive leadership team. Tony Gibney is stepping down from his position as chief financial and business officer, while Conor Kilroy is leaving his role as chief legal officer. Mark Plavsic has also departed as chief technology officer.

At the same time, Aura has appointed Susan Abu-Absi as chief operating officer, Erica Kratz as chief regulatory and quality officer, and Julie Person as chief people officer. An existing employee will serve as interim chief financial officer.

Alongside the operational changes, Aura projects that its revised spending plans will extend its cash runway into the first half of 2029, compared with the second half of 2028 under its previous plan.
