Dimerix has acquired DMX-652, a phase 2-ready drug candidate for acute kidney injury (AKI), from Mission Therapeutics in a deal that includes a $5 million upfront payment and potential milestone payments of up to $287 million.
The acquired asset, previously known as MTX652, is a selective inhibitor of USP30, a mitochondrial enzyme that slows the removal of damaged mitochondria. Mission Therapeutics developed the molecule and generated phase 1 data in healthy volunteers before receiving approval in 2023 to proceed into phase 2 development. The company later shifted its focus to MTX325, a Parkinson’s disease program, and divested the AKI asset.
Under the agreement, Dimerix could pay up to $47 million in clinical development milestones, $40 million upon marketing approval, and $25 million if the drug is approved in a second indication. The remaining $175 million is linked to sales milestones.
The acquisition includes assignment of the composition of matter patent, an open U.S. Investigational New Drug (IND) application, a phase 2 clinical trial protocol that has received approval from the U.S. Food and Drug Administration to proceed, manufacturing methodology, and sufficient pharmaceutical-grade drug product for the planned study.
Dimerix has renamed the asset DMX-652 and plans to initially focus development on AKI. The phase 2 trial is designed to investigate the drug’s potential to prevent kidney injury and preserve renal function. The company stated that the mechanism of action may also allow the candidate to be developed in additional indications.
Mission’s phase 1 clinical trial enrolled 85 healthy volunteers. The study found that DMX-652 was safe and well tolerated when administered as single doses of up to 200 mg per day and multiple doses of up to 100 mg per day over 14 days. No serious adverse events related to the drug were reported, and the trial generated pharmacokinetic data supporting further development.
According to information provided by Mission, preclinical studies suggested that inhibiting USP30 could protect against tubular atrophy and fibrosis by promoting the removal of damaged mitochondria. The drug is intended to support mitochondrial quality control in injured kidney cells and is being investigated for AKI associated with ischaemia, toxins, or sepsis-related causes.
The acquisition expands Dimerix’s kidney disease pipeline. The company’s lead candidate, DMX-200, is currently being evaluated in a fully recruited phase 3 trial in focal segmental glomerulosclerosis (FSGS), a rare kidney disease. Dimerix has stated that DMX-652 complements its existing renal-focused portfolio and broadens its activities from glomerular disease and FSGS into acute kidney injury.
Commenting on the transaction, Dimerix Chief Executive Officer and Managing Director Dr. Nina Webster said, “DMX-652 represents an exciting and differentiated novel compound, and the acquisition of a Phase 2-ready program in acute kidney disease represents an important step in executing our strategy to expand our renal pipeline.”
For Mission Therapeutics, the transaction provides non-dilutive funding as it advances MTX325, another USP30 inhibitor. Mission began a phase 1 trial of MTX325 in healthy volunteers and people with Parkinson’s disease in 2023. After completing phase 1a testing and raising $13.3 million, the company committed to advancing the program into a phase 1b proof-of-mechanism study in patients with Parkinson’s disease.
